MIRI® Time-Lapse Incubator

The MIRI® Time-Lapse Incubator is a multiroom incubator with a built-in camera and microscope. Designed and manufactured in EU, the MIRI® Time-Lapse Incubator provides high quality time-lapse images of embryos developing in "real-time" without having to remove the embryos from the safety of the incubation chamber for manual microscopy. Time-lapse embryo monitoring provides detailed morphokinetic data throughout embryo development, which is not available on routine spot microscopic evaluation. This allows all important events to be observed, helping to identify healthy embryos with the highest probability of implantation, with the aim of achieving higher pregnancy rates.

Addressing IVF Risks

Time-lapse technology is a modern system used in the practice of Clinical Embryology for the culture and imaging of embryos in vitro. In recent years, the risks associated with In Vitro Fertilization (IVF), such as multiple pregnancies and higher rates of mother and neonate mortality, have increased.

Although stricter policies on embryo transfer have reduced some of the risks involved, the occurrence of twins is still a concern. It is believed that a twin pregnancy can be avoided by opting for a single embryo transfer. However, the traditional method of selecting embryos based on their morphology is not accurate enough to allow for routine single transfers. Thus, new and more effective screening tools are needed.

By utilizing a time-lapse incubator, it is possible to continuously monitor embryos without disturbing their optimal culture conditions. This non-invasive method enables IVF practitioners to observe important information such as cleavage patterns, morphological deviations, and the dynamics of embryo development. Constant monitoring can aid in identifying embryos with higher implantation potential, allowing for unbiased selection of embryos for transfer. This technological advancement is a significant step forward in the field of IVF.

The Silent Embryo Hypothesis

Baumann et al wrote that “The Silent Embryo hypothesis proposes that viable preimplantation embryos operate at metabolite or nutrient turnover rates distributed within lower ranges than those of their less viable counterparts”. 

The MIRI® TL Multiroom IVF incubator is a multi-room incubator equipped with a built-in microscope and camera that continuously captures images of embryo development without disruption until the day of transfer. This allows for a detailed scoring of the cultured embryos, which can better predict their developmental and implantation potential. 

In conventional assessment of embryo selection in IVF, observations are often limited to pre-defined time intervals, which requires removing the embryo from the incubator. With the MIRI® TL Multiroom IVF incubator, the dynamic process of embryo development can be observed without disturbing the culture conditions. This supports the Silent Embryo Hypothesis, which suggests that embryos develop better when they are less disturbed.

Simple guide on how to operate the MIRI® TL

I. CultureCoin® Preparation

  1. Leave the dishes with the covers on for off-gassing in the aseptic environment for 12 hours (overnight).
  2. Warm the dishes on a heated stage or an incubator before filling them with media.
  3. Fill all the culture wells with approximately 25 µl of precultured equilibrated culture media in an aseptic environment.
  4. Filling of the washing wells is optional. One washing well contains approximately 23 µl of the solution. They can be used for cell manipulations, washing media or replacement media.
  5. Check under a microscope for any visible air bubbles and remove them with a stripper tip. 
  6. Fill the oil area with a confluent layer of suitable mineral or Paraffin oil to cover the culture media. The recommended amount is 6 ml. Do not overfill. 
  7. Put the cover on the dish and leave it for 4 hours of equilibration in the CO2 incubator (humidified incubator is preferable).

II. Loading of the Embryos

  1. Check for air bubbles in the culture wells after the equilibration. Remove any bubbles with a stripper tip. 
  2. Load the embryos in the culture wells. Make sure the embryos are located very precisely in the center of the culturing well and not on the side slope.
  3. Optionally fill the pH well with culture media up to the lowest step (recommended volume – 350 µl) of the pH well and cover it with an oil overlay (recommended volume – 150 µl).
  4. Once done, put the CultureCoin® in the MIRI® TL family’s multiroom IVF incubator.
  5. Select the patient, then select or deselect desired active positions and start the time-lapse when ready.

III. Embryo Annotation

  1. On the MIRI® TL Viewer's Home Screen, select “Timelapses” and look for the patient to open time-lapse videos. A revolver view with the time-lapse video of each of the 14 wells in the CultureCoin® will be displayed.
  2. Select an embryo to annotate. The embryo will be maximized at the center. The annotation system is structured around the “events” that are located on the left of the wheel.

IV. Selecting the most Viable Embryos

  1. The MIRI® TL Viewer software is equipped with sophisticated tools to help you evaluate and select the most viable embryo after completely annotating all embryos.
  2. The Compare Tool will let you do a side-by-side comparison of two embryos. Two embryo videos can be played independently or linked together for comparison.
  3. The “Summary” tool will allow you to compare all embryos in an informative and graphical way. It will let you compare embryo developmental timings and which one develops closely with the ideal timings.
  4. Once the selection process has been completed or a decision has been made, click the green check icon in the upper left corner. After clicking it, a drop-down list of the desired status will appear.
  5. Assign a corresponding color that indicates the decision in each well: Green for “transfer”, Red for “discard”, and Blue for “freeze”. The colored ring will appear around the well to indicate the decision made. You can also click the “Dish Map” icon to visually map the decision that has been made.
